Immune checkpoint blockade (ICB) immunotherapy delivers promising clinical results in colorectal cancer (CRC). However, few CRC patients are eligible for and respond to current ICB targets. Therefore, there is a need to identify other checkpoint targets associated with the tumor microenvironment and uncover their mechanism of action. Our findings suggest that tumoral ST2 signaling drives immune escape through a reduced antigen presentation and could serve as checkpoint target for CRC immunotherapy.
