This lecture series showcases the exceptional work being done by early-career faculty members and provides a unique opportunity for informal critique and scientific discussion amongst junior and senior faculty members. Benjamin Diamond, MD, Assistant Professor of Medicine, Division of Myeloma, and Sylvester K12 graduate, will discuss his work on recurrent genomic resistance mechanisms and relapse patterns in hematologic malignancies. Current immunotherapeutics in lymphoid malignancies target cell-surface antigens. While monoclonal antibodies and T-cell redirecting therapies have revolutionized care, many patients suffer inevitable relapse of disease. We explore the genomic loss or alteration of cell-surface targets as a recurrent mechanism of resistance to anti-CD38 antibodies and BCMA- and GPRC5D-directed T-cell engagers. Using chemotherapy and radiotherapy-related mutational signatures, we can time the acquisition of these events thoughout a patient’s treatment journey.
