Clinical trials testing new treatments are divided into different stages called phases. The primary objective of Phase I trial design is to determine dose and schedule, and the primary endpoints are safety, pharmacokinetics, toxicity profile, and modulation of target/biomarker, all to help guide later phase clinical trials. This lecture will discuss the goal of Phase I studies in oncology and provide an introduction to Phase I study designs. Rule-based, model-based, and model-assisted designs will be presented.
