Advanced Imaging Modalities and Applications for Cancer Research: Expanding Capabilities and Services in the Flow Cytometry Shared Resource

https://miami.zoom.us/webinar/register/WN_e44P-7t6T9-oGsG2_z2C4QThe Sylvester Flow Cytometry Shared Resource (FCSR) offers state-of-the-art instruments and services that are critical to the success of a wide range of cancer research. This seminar will highlight the expansion of the FCSR’s advanced imaging services and provide case studies. The FCSR offers a Zeiss LSM 980 laser scanning confocal microscope with Airyscan 2 that provides low phototoxicity and high speed; simultaneous spectral detection of multiple weak labels with high-light efficiency; a wealth of fluorescent labels; and spectral flexibility with simultaneous channels. This microscope provides confocal imaging with 3D optical sectioning and reconstruction; super-resolution down to 90 nm; wide-field, multi-well plate imaging; tile scanning of large regions; multispectral detection for emission fingerprinting and spectral unmixing of markers; calcium imaging; and an AI-enabled imaging area finder. Moreover, this microscope provides time-lapse, live cell imaging over hours or days (e.g., 72+ hours) with environmental regulation. The FCSR also offers the Standard Biotools CyTOF Hyperion imaging mass cytometer, which utilizes heavy metal-tagged antibodies, immunolabeling, and time-of-flight mass spectrometry to measure up to 50 protein markers in FFPE or frozen tissue sections (or fixed cells adhered to a slide) at 1 um resolution, enabling spatial proteomics imaging and characterization of tumor microenvironments at single-cell resolution. The FCSR has a repository of validated antibodies-to-human-markers and a heavy metal-antibody conjugation service to facilitate use of this system.
